Cloning, sequencing and expression of the prn pertactin gene of canine origin Bordetella bronchiseptica

Abstract
PERTACTIN (PRN) IS AN OUTER MEMBRANE PROTEIN OF BORDETELLA BRONCHISEPTICA, IT HAS ADHESIVE PROPERTIES AND THE AUTO-TRANSPORTER PROPERTIES OF THE TYPE V SECRETION SYSTEM. THIS PROTEIN HAS TWO VARIABLE REGIONS IN THE PRIMARY STRUCTURE, WHICH INDUCES PROTECTIVE IMMUNITY IN PIGS AND MICE. IN THIS STUDY, THE PRN GENE OF B. BRONCHISEPTICA (P98 WILD TYPE) OF CANINE ORIGIN WAS CLONED, SEQUENCED AND EXPRESSED IN E COLI. PRN WAS DETECTED BY IMMUNOBLOT USING ANTIBODIES AGAINST BORDETELLA. THE RGD DOMAIN WAS IDENTIFIED DUE TO THE SEQUENCE OF AMINOACIDS, WHICH IS THE EPITHELIAL CELL ADHERENCE DOMAIN BETWEEN 261 AND 263 AMINOACIDS AND TWO REPEAT REGIONS BETWEEN GLY 264 AND PRO 278 THAT CORRESPOND TO THE REGION I AND PRO 568 TO PRO 587 FOR THE REGION II.
